apoptosis: Acridine Orange - Eth Br

I have read a great
deal about apoptosis and I had come to the conclussion that a conformatory
DNA gel would be essential. I however thought that it would be nice if I could
use the flowcytometer to detect apoptosis as a quick and easy way. The whole
idea originated from a seminar here at our institute where such a technique
was discussed. {This was by a french researcher named Marie-Francois from
Pastuer Institute} I am unable to find a published paper for this techiniqe
but essentially it uses Acridine Orange and Etidium Bromide together to
detect apoptosis. Since my project is not directly involved with apoptosis and
as my only interest is to see if a factor I have causes Immunosuppression by
apoptosis, I was keen on geting this protocol. The principle is the same as
that discussed in Current Protocols in Immunlogy but I need some additional
info on where my particular population apoptotic/non-apoptotic would apear
on a double parameter histogram of Acridine orange intensity vs Ethidium bromide
intensity. I would appreciate your comments and suggestions on this issue.
