Regarding staining yeast cells for DNA analysis, you probably have to digest
away the cell wall with an enzyme such as glusulase before you do the
standard fixation. Yeast cells are not like mammalian cells in that they
have a cell wall as well as a cell membrane. I have not actually done DNA
analysis in yeast, however, so hopefully someone with experience doing this
will answer your query. In any case, fixation alone is unlikely to do what
you need.
