Ed: you have just asked a very important question. I would be most
interested in seeing the various portocols used in the US, UK and Europe.
As you probably know there is tremendous variation between labs. I am
interested in testing various protocols in an attempt to find a standarized
method(s). I just returned from the 3rd European Stem Cell Methodology
Workshop in Mulhouse, France. This group is conducting a multicenter study
of the flow cytometric determination of CD34+ cells. A couple of studies
have been completed in the US. And I am sure that there will be more
recommendations/suggestions. Any and all comments regarding the flow
cytometric standardization of CD34+ cells and CD34+ subsets will be
welcomed. I am trying to prepare an in house review on this subject.

How do people out there measure the CD34+ population by flow, do you
>gate on CD45+ cells or around the MNC fractions?
