Monocyte sorting

We are interested in sorting viable human monocytes (CD14+) to be used in
functional cytokine production assays. We have done the experiment once
with poor monocyte viability. The monocytes were sorted, placed into
culture for 24 hours and then stimulated with LPS and monensin. The
combination of these events killed the monocytes - any suggestions? Thanks
